The Investigation of Callogenesis and Somatic Embryogenesis of Strawberry

Abstract

Callogenesis and somatic embryogenesis of strawberry explants was investigated in vitro. To do this, strawberry explants cultured on MS medium that modified with combination of plant growth regulators. For statical analysis, the callogenesis was established with 2 factors (hormone and organ) in a complete randomized block design in 4 replications and somatic embryogenesis was analyzed with t test between petal and receptacle. In this experiment, explants infection and browning were the main problems of strawberries micro propagation. Results showed that callus initiation was observed in strawberry leaf and petiole on MS medium that supplemented with TDZ (12 µM) + IBA (0.05 µM) and BAP (44.4 µM) + NAA (0.05 µM). Callus fresh weight was significantly different between 2 organ and hormone component, but there was no difference in callus dry weight (p>0.05). Histological experiment clears that cambium layer produce callus in strawberry petiole. Petals and receptacles was highly responsive with regard to embryo induction in TDZ (12 µM) + IBA (0.05 µM). Petal explants significantly gave the maximum number of somatic embryos to compare of receptacle embryos.
