تولید فورسکولین و بیان ژن 1- دی‌اکسی-D-زایلولوز-5- فسفات ردوکتوایزومراز در گیاه حسن‌یوسف (Coleus forskohlii) تیمار شده با مس

چکیده

فورسکولین، ماده مؤثره گیاه Coleus forskohlii است که خواص درمانی متعددی دارد. در این تحقیق، میزان تولید فورسکولین و همچنین بیان ژن آنزیم 1-دی‌اکسی-D- زایلولوز-5- فسفات ردوکتوایزومراز (DXR) در گیاه C. forskohlii تیمار شده با غلظت‌های مختلف (صفر (به­عنوان گیاه شاهد)، 4، 8 و 16 میکرومولار) الیسیتور مس به‌مدت 16 ساعت بررسی گردید. نتایج نشان‌دهنده افزایش تولید فورسکولین تا غلظت 8 میکرومولار است، به­طوری­که در این تیمار، مقدار فورسکولین حدود 5/1 برابر مقدار آن در نمونه شاهد افزایش یافت که با تغییرات سطح بیان ژن DXR همخوانی داشت. ثابت شده است که حضور اضافی مس باعث تولید مولکول‌های فعال اکسیژن می‌شود که به­طور مستقیم و یا با واسطه مولکول‌های سیگنالینگ دیگر، بیان ژن‌های درگیر در سیستم دفاعی گیاه را فعال می‌کنند. با توجه به نقش دفاعی دی‌ترپن‌ها، احتمال داده می‌شود نتیجه افزایش بیان ژن DXR تحت تیمار با این الیسیتور، افزایش تولید فورسکولین است که نقش محافظتی در برابر تنش‌های را داراست.

Forskolin is an effective compound found in Coleus forskohlii plant which has different therapeutic properties. In this research, production of forskolin and gene expression of 1-deoxy-D-xylulose-5-phosphate reductoisomerase (DXR) were investigated in C. forskohliiplant after treatment with different concentrations of copper elicitor (0; as control, 4, 8 and 16 µM) for 16 hours. Results showed that forskolin concentration is increased up to 8 µM in such a way that the concentration can be estimated 1.5 fold more than the control sample, which consisted with changes of DXR gene expression level. It has been proved that excess concentration of copper, causes triggering of reactive oxygen species that directly or mediated through others signaling molecules induce gene expression level of defensive system. Considering the defensive role of diterpens, it is proposed that increase in DXR gene expression level by copper elicitor leads to increase in forskolin concentration as protective compound against stresses.
