واکنش گیاهچه¬های حاصل از کشت بافت سیب¬زمینی به نیتروژن و اسید جاسمونیک در شرایط هیدروپونیک

چکیده

واکنش گیاهچه­های سیب­زمینی (رقم مارفونا) حاصل از کشت بافت به افزودن نیتروژن و اسید جاسمونیک در تولید ریزغده در شرایط هیدروپونیک آزمایشی به­صورت فاکتوریل در قالب طرح کاملا تصادفی در گلخانه تحقیقاتی مورد بررسی قرار گرفت. تیمارهای آزمایش شامل نیتروژن در سه سطح صفر (شاهد)، 50 و 100 میلی­گرم در لیتر محیط کشت، از منبع نیترات آمونیم و اسید جاسمونیک به­صورت محلول­پاشی بر روی برگ­ها در چهار سطح صفر (آب مقطر به­عنوان شاهد)، 1، 5 و 10 میکرومولار در سه تکرار بودند. شاخص­های اندازه­گیری شده عبارت بودند از: تعداد و وزن کل ریزغده­ها، متوسط وزن ریز غده، ارتفاع بوته، تعداد برگ و شاخص برداشت. نتایج نشان داد که بیش­ترین تعداد ریزغده، وزن کل ریزغده، متوسط وزن ریزغده و شاخص برداشت از مصرف 50 میلی­گرم نیتروژن توام با 5 میکرومولار اسید جاسمونیک به­دست آمد. هم­چنین بالاترین ارتفاع ساقه و تعداد برگ به­ترتیب متعلق به تیمارهای 50 میلی­گرم نیتروژن بدون جاسمونیک اسید و 100 میلی­گرم نیتروژن توام با یک میکرومولار اسید جاسمونیک بود. کم­ترین تعداد و وزن کل ریزغده و تعداد برگ از محلول­پاشی 1 میکرومولار اسید جاسمونیک بدون مصرف نیتروژن به­دست آمد.

چکیده [English]

The response of potato plantles derived from tissue culture to nitrogen and jasmonic acid adding under hydroponic condition was studied in a factorial experiment based on a complete randomized design. The study was carried out in a research green house.  Experimental treatments included 3 levels of nitrogen (0, 50 and 100 mg/l of culture media) from the source of ammonium nitrate and 4 levels of jasmonic acid (0, 1, 5 and 10 µM) in three replicates. Total number and weigh of minitubers; average weight of minituber; plant height and number of leaves; and harvest index were studied. The results showed that the highest total number and weight of minituber, average weight of minituber, and harvest index obtained from the treatment of 50 mg nitrogen plus 5 µM jasmonic acid. Also the highest plant stem and leaf number obtained from the treatments of 50 mg nitrogen without jasmonic acid and 100 mg nitrogen plus 1 µM jasmonic acid respectively. The lowest number and total weight of minituber and leaf number; belonged to the treatment of 1 µ M jasmonic acid without nitrogen.
